Im mounting my calipers on plates all as per the Manky ones axle. But I can see  that moving the mounting plate so that the caliper will be radially 90 degrees forward,ie in front of the axle centre line instead of behind the axle line ,I can get a better h/brake cable run. Will this be detrimental to braking efficiency, due to being 'leading' as opposed to 'trailing'?

Leading/trailing brake shoes are more to do with the mechanism operating the brake shoe rather than the orientation. (Lining length, thickness and position can be different on a pair of shoes.)

Link below explains better.

With disc pad system it makes no difference where the pads “clamp” the disc surface.

yep, caliper position will make no difference to the brake performance, however, think about where the bleed screw will end up.
